ABSTRACT

Objective To investigate the effects of Qibai Pingfei Capsules(QPC) on the expression of monoamine neurotransmitters of 5-hydroxytryptamine receptor(5-HT) and dopamine(DA) receptors of DA1 and DA5 in the rats with chronic obstructive pulmonary disease (COPD) accompanied with depression, and to explore the possible therapeutic mechanism. Methods Ninety male Wistar rats were randomly divided into 6 groups, namely normal group, COPD depression group, QPC group, Venlafaxine group, and QPC plus Venlafaxine group. The rat model of COPD accompanied with depression was established by compound methods of smoking, food deprivation, water deprivation, forced swimming, shaking, and clipping tail. And then the effects of QPC on the lung function and the general characteristics were observed. Furthermore, the mRNA level of 5-HT1AR was determined by real-time quantitative reverse transcription PCR(RT-qPCR), and protein levels of DA-1 and DA-5 in rat hippocampus were assayed by Western blot analysis. Results Compared with the normal group, forced expiratory volume in 0.3 second(FEV0.3), the forced volume capacity(FVC) and FEV0.3/ FVC in the COPD depression group were significantly reduced (P < 0.01). Compared with the COPD depression group, the values mentioned above were restored to different extents in the groups of QPC, Venlafaxine, and QPC plus Venlafaxine(P<0.01). Compared with the normal group, the expression levels of 5-HT1AR mRNA, and DA-1 and DA-5 protein in the COPD depression group were significantly down-regulated, and their levels were also slightly decreased in the treatment groups(P<0.05). Compared with the COPD depression group, the expression levels were up-regulated to various degrees in all of the treatment groups (P < 0.05). Conclusion QPC can increase the mRNA levels of 5-HT1AR and protein levels of DA-1 and DA-5 in the hippocampus of COPD with depression.

ABSTRACT

Objective To investigate the effect of Chaigan Jieyou Decoction on hippocampal neurons and the expression of hippocampal brain-derived neurotrophic factor(BDNF)in rats of Parkinson’s disease with depression (PDD). Methods Twenty-four adult male SD rats were randomly divided into 4 groups, namely normal group, model group, Chaigan Jieyou Decoction group and Fluoxetine group, 6 rats in each group. Except for the normal group, the rats in the other groups were fed solitarily. After the establishment of PDD model, the model group, Chaigan Jieyou Decoction group and Fluoxetine group were given intragastric administration of distilled water 2.0 mL, Chaigan Jieyou Decoction 8 g·kg-1, Fluoxetine 1.8 mg·kg-1 for one day respectively, once a day. The treatment covered 21 continuous days. The hippocampal neuron damage and the expression of BDNF in hippocampal tissues were detected by Nissl’s staining and immunohistochemistry, respectively. Results Compared with the normal group, model group had obvious morphological damage of the hippocampal neurons, while Chaigan Jieyou Decoction and fluoxetine could relieve the damage, showing certain protection of hippocampal neurons. Compared with the normal group, BDNF positive staining area, integral optical density (IOD), and mean optical density(MOD) of the hippocampus in the model group were significantly decreased (P<0.01). Compared with the model group, BDNF positive staining area, integral optical density and mean optical density in Chaigan Jieyou Decoction group and fluoxetine group were significantly increased(P<0.05 or P<0.01). Conclusion Chaigan Jieyou Decoction has an effect on the relief of the damage of neurons and the increase of expression of BDNF in hippocampus of rats with PDD, which probably contributes to one of its therapeutic mechanisms.

ABSTRACT

Objective To investigate the effect of β-asarone on hippocampal neurons of experimental depression rats. Methods Eighty male adult SD rats were randomly divided into 4 groups, namely normal group, model group, fluoxetine(1.2 mg/kg) group and β-asarone (25 mg/kg) group. Depression rat model was established. The medication groups were given corresponding agents respectively. After treatment for 21 days, the number of apoptotic cells and the histological features in the hippocampus of rats were detected by flow cytometry and TUNEL, respectively. Results Compared with the normal group, typical apoptotic cells were found, apoptotic cell count and apoptotic rate were increased in the hippocampal CA3 and DG area of the model group (P<0.01). Compared with the model group, apoptotic cell count and apoptotic rate were decreased in the hippocampal area ofβ-asarone group and fluoxetine group (P<0.01). Conclusion The protective mechanism of β-asarone for the hippocampal cells of depression rats is probably associated with the reduction of the apoptosis of hippocampal neurons.

ABSTRACT

【Objective】To observe the anti-depression effect of Shugan Hewei Granules(SHG) on mice with behavioral despair.【Methods】Kunming mice were randomized into 5 groups: model group,low-,moderate-and high-dose SHG groups(in the dosages of 0.25,0.5 and 1.0 g?kg-1?d-1respectively) and chloropromazine(CP) group.Tail suspension test and forced swimming test were used to investigate immobility time of mice.【Results】Moderate-and high-dose SHG shortened the immobility time obviously during tail suspension test and forced swimming test(P

ABSTRACT

【Objective】To investigate the antianxietic action of Xiaoyao San(XS)and Dan Zhi Xiaoyao San(DZXS).【Methods】 Wistar rats were randomized into 5 groups: model control group,high-and low-dose DZXS groups(14.4?g/kg and 7.2?g/kg respectively),and high-and low-dose XS groups(21.06?g/kg and 10.53g/kg respectively).Social interaction test and open-field test were carried out to observe the antianxietic action of XS and DZXS.Except that the model group was given normal saline, the rats in other groups were given the corresponding drugs according to the experimental design for 14 successive days. After treatment, the activities within 5 min were monitored by two observers and then the mean value were calculated.【Results】High-and low-dose DZXS prolonged rats social interaction time and increased the times of body erection and dressing their hair, and low-dose XS also increased the times of body erection and dressing their hair(P
